DEFINITION

Haptoglobin (406 aa, ~45 kDa) is encoded by the human HP gene. This protein plays a role in tight junction disassembly, intestinal permeability, heme iron recycling, antimicrobial activity and the regulation of hydrogen peroxide catabolism.

Binding free plasma hemoglobin, secreted haptoglobin prevents loss of iron through the kidneys, protects the kidneys from oxidative damage by hemoglobin (Hb), and makes hemoglobin accessible to degradative enzymes. Hp/Hb clearance appears to be mediated by the monocyte/macrophage scavenger receptor CD163. Uncleaved haptoglogin, also known as zonulin, plays a role in intestinal permeability, by allowing intercellular tight junction disassembly, and controlling the equilibrium between tolerance and immunity to non-self antigens.
