DEFINITION

Prothrombin (622 aa, ~70 kDa) is encoded by the human F2 gene. This protein plays a role in both coagulation and proteolysis of fibrinogen.

Prior to secretion, prothrombin is modified to have gamma-carboxyglutamyl residues, resulting from the carboxylation of glutamyl residues by vitamin K-dependent gamma-carboxylase. These modified residues are necessary for the calcium-dependent interaction with a negatively charged phospholipid surface, which is essential for the conversion of prothrombin to thrombin.
