| DEFINITION |
Prothrombin (622 aa, ~70 kDa) is encoded by the human F2 gene. This protein plays a role in both coagulation and proteolysis of fibrinogen. |
| DesignNote |
Prothrombin protein can be cleaved into the following 4 chains: activation peptide fragment 1, activation peptide fragment 2, thrombin light chain, thrombin heavy chain. (UniProt) Prothrombin activates factors V, VII, VIII, XIII, and, in complex with thrombomodulin, protein C. (UniProt) Prothrombin catalyzes the cleavage of Arg-Gly bonds in fibrinogen to form fibrin and release fibrinopeptides A and B. (UniProt) Prior to secretion, prothrombin is modified to have gamma-carboxyglutamyl residues, resulting from the carboxylation of glutamyl residues by vitamin K-dependent gamma-carboxylase. These modified residues are necessary for the calcium-dependent interaction with a negatively charged phospholipid surface, which is essential for the conversion of prothrombin to thrombin.
